Meis proteins belong to the TALE-subfamily of homeodomain containing transcription factors and play multiple roles in development and disease. They form multimeric complexes with other proteins to control gene expression. Different proteins have been shown to interact with Meis family members in non-neuronal tissue and in the posterior central nervous system (CNS). Meis-interacting proteins in the anterior CNS have remained elusive. As we recently noticed, the Meis family protein Meis2 is expressed in the subventricular zone of the lateral ventricle (SVZ) and the rostral migratory steam (RMS). The SVZ is one of the major sites in the adult mammalian brain, where new neurons are generated throughout life. Meis2 function in this important brain region is entirely unexplored at present. Here I propose to investigate the physiological role of Meis2 during adult neurogenesis in the SVZ/RMS system by two complementary experimental approaches: (i) the transfection of individual cells in the SVZ and RMS with retroviruses that carry different forms of Meis2 lo investigate Meis2 function during adult SVZ neurogenesis and (ii) the biochemical purification of Meis2 interacting proteins in the SVZ and RMS. This research program will yield new insights into the function of Meis proteins in the CNS and presents a novel approach to decipher the genetic program that controls neurogenesis in the adult mammalian brain.
