The tedanolides and candidaspongiolides represent a unique class of secondary metabolites that are highly potent inhibitors of eukaryotic protein biosynthesis in the sub-nanomolar range. Structurally, this macro lactone made from a primary alcohol is an unrivaled motif. Since this is not only a rare structural element but also mechanistically not in accordance with the pivotal thioesterase reaction in polyketide biosynthesis, detailed studies of the mechanism of this macrolacton formation will provide insight in the synthesis of this class of natural products and at the same time pave the way for a detailed understanding of the interplay between structural motifs and their biological consequences.
