During the two previous funding periods our work has established quantitative shotgun lipidomics by high-resolution mass spectrometry as a generic analytical tool applicable to a variety of problems in lipid biology and medicine from high-throughput clinical screens to targeted quantification of individual lipids within lipid-protein assemblies. However, the expanding application base together with steadily increasing performance of Orbitrap mass spectrometers presents new challenges for the shotgun methodology. Within the next period we propose to focus on three key aspects of high resolution shotgun lipidomics: efficient processing of thousands of high resolution shotgun FT MS/MS spectra; accurate quantification of lipids with polyunsaturated and / or labile fatty acid moieties and quantifying lipid flux at the uncompromised lipidome coverage and sensitivity. The anticipated developments will make sure that shotgun methodology is not overpowered by the richness of high resolution spectra, but is taking full advantage of increasing mass accuracy and resolution and achieves more precise, comprehensive and dynamic characterization of lipidomes.
