Recently we showed that the priming factor CAPS1 supports synaptic transmission of dorsal root ganglion (DRG) neurons while its paralog CAPS2 promotes the release of neuropeptides at their somata. These functions of both CAPS paralogs are based on their different subcellular localizations. In the next funding period we will: 1. Investigate the underlying molecular mechanism of CAPS1 and CAPS2 localization in DRG neurons; 2. Determine the difference of CAPS-dependent neuropeptide release between soma and synapses; 3. Investigate the role of CAPS2 in the development of neuropathic pain in animals.
