Project P11 focuses on the role of Rab5-like Vps21 and Rab7-like Ypt7 in coordinating endosomal maturation and fusion at endosomes and vacuoles. In the next period, the group will take advantage of its purified components and novel assays to further dissect and reconstitute sub-reactions of endosomal maturation. This will include (i) the analysis of the requirement for successful and efficient transition from Vps21 to Ypt7, (ii) the detailed characterization of Retromer function in cargo-mediated activation on endosomes and vacuoles, in particular in comparison to the AP-3 complex, and (iii) the coordination of the endosomal Rabs with tethering complexes during endosomal and vacuole tethering.
