Targeting the very short and short living structures of the various intermediates formed in reversibly photoswitchable fluorescent proteins during their photo life cycle is the major aim of project B10. Technically, we will face this ambitious goal by applying novel and state of the art time-resolved x-ray scattering and time-resolved x-ray diffraction experiments at synchrotron and FEL sources. We will utilize the structural dynamics data to develop novel and optimized variants of reversibly photoswitchable fluorescent proteins for applications in super-resolution microscopy.
