We are planning to explore the effect of pressure modulation on the spatio-temporal patterns and reaction dynamics of membrane-associated signaling proteins of the Ras family using static and dynamic spectroscopic and microscopic techniques. In a first step, we plan to start with a minimum Ras signaling machinery using N-Ras and K-Ras and the presence of selected regulators and effectors (such as GTP/GDP, Sos, Raf) to explore the effect of pressure perturbation on the membrane partitioning, lateral organisation, and the reaction dynamics of the membrane-associated components. Based on these results on reconstituted membrane systems, we will then - in a later stage of the project - extend the use of the pressure perturbation approach for the modulation of reaction networks and identification of protein connectivity patterns also to in vivo systems.
