Time resolved fluorescence detection utilising two photon excitation in microfluidic separation systems

This project focuses on the development of a novel approach for label-free detection of tiny sample amounts in chip-based separation systems. This will be achieved by two photon fluorescence excitation combined with time resolved fluorescence detection of the emission light. In comparison to common single photon excitation in the deep UV-spectral region, this should enable label free detection in economic chip materials which are not transparent below 300nm, such as borosilicate glass or plastics. On the fly determination of fluorescence life times will be utilized for reliable peak assignment in chip electrophoresis. The potential of two photon excitation in microfluidic systems will be evaluated systematically with of a high performance system for fluorescence excitation and detection. Finally the developed systems and methods will be evaluated in several application areas, such as Bionanalytics, drug analysis and integrated organic synthesis.
