TP09 investigates how the response of cells to proteotoxic stress is regulated at a molecular level. One focus of the project is the analysis of the structure of monomeric and trimeric human heat shock transcription factor Hsf1 and the development of an Hsf1 based bioluminescence resonance energy transfer sensor that could be used to screen for Hsf1 activating and Hsf1 inhibiting small molecules as therapeutics for cancer and neurodegenerative diseases. The second focus is the molecular mechanism of the modulation of Hsf1 activation and attenuation by posttranslational modifications and interacting proteins.
