Obesity is associated with enhanced severity and risk of chronic inflammatory diseases such as psoriasis, multiple sclerosis, asthma, rheumatoid arthritis as well as with a reduced therapeutic success in these diseases. However, mechanistic links between alterations in adipose tissues and the dysregulation of immune responses in obesity-associated chronic inflammatory diseases are poorly understood. Project B5 tests the hypothesis that alterations of adipose tissue in obesity are responsible for the obesity-mediated exacerbation of chronic inflammatory diseases, exemplified in the signature disease psoriasis. We also ask whether central players in the pathogenesis of psoriasis, such as TNFalpha, IL-1beta, IL-17, are involved in the functional control of adipose tissue thus supporting the development of obesity.
