Hsp47 is essential for proper collagen folding and secretion and its upregulation under fibrotic conditions makes it a candidate target for anti-fibrotic drugs. We have characterized its client repertoire and will explore its chaperoning of different clients further. We have studied in detail its mechanism of pH-dependent client binding and release. To investigate its role in normal skin development and fibrosis we have created fibroblast and keratinocyte-specific knockout mouse lines. With potential inhibitors identified in a high-throughput screen we will explore Hsp47’s therapeutic potential in mouse models with skin fibrosis.
