Aim of the project A08 is the functional characterization of the human-relevant NPSR1 polymorphism (N107I). An analogous mutation (I107N) has been introduced into a new mouse model using CRISPR/Cas9-mediated gene editing. The functional consequence of this mutation on neuronal properties, on neuronal circuits in fear relevant brain regions (e.g. amygdala, intralaminar thalamus), fear extinction and generalization will be analyzed in vitro/ex vivo using electrophysiological and optogenetic techniques and in vivo using established behavioral- and stress-paradigms.
