Our project explores potentially lipid-modulating mechanisms in the progression of hepatic inflammation, injury and fibrosis in chronic non-alcoholic steatohepatitis (NASH) and acute alcoholic steatohepatitis (ASH). Specifically, we will study the hypotheses (1) that, in patients with NASH, specific processing of nutrients produces a disease-specific postprandial plasma lipidome that is relevant for NASH progression, (2) that upregulation of lipoprotein lipase, which is characteristic for NASH, is a compensatory mechanism to inhibit NASH progression and (3) that pharmacological inhibition or genetic deletion of myeloperoxidase attenuates ASH.
