The identity and function of organelles is determined by their protein and lipid composition. Recently, it became obvious that both unfolded proteins and aberrant lipid compositions of the endoplasmic reticulum (ER) membrane are potent activators of a homeostatic program, the unfolded proteins response (UPR). We are studying the most conserved transducer of the UPR, Ire1, and have identified an amphipathic helix that is capable to sense aberrant lipid compositions. Now, we aim at identifying the activation mechanism of Ire1 by lipid bilayer stress in order to define the term "ER-stress" at the molecular level.
