We apply advanced microscopic techniques to characterize critical steps in early HIV-1 replication on the single particle level with high spatial resolution in physiologically relevant cell systems. We have shown that apparently intact capsids enter the nucleus through intact nuclear pores in all cell types analyzed, and un-coating of the viral genome occurs by rupture of the capsid inside the nucleus. Building on these observa-tions, which have changed the general view of early HIV-1 replication, we will interrogate the hypothesis that intact capsids orchestrate the entire early phase of HIV-1 replication. Furthermore, we will investigate the process of genome uncoating in the nucleus by rupture of the capsid lattice.
