This grant application is for a cell sorter to replace a 17-year-old high speed cell sorter. The cell sorter company will no longer offer service or support for the machine after the end of 2013, making it urgent that we find a replacement which is technically up-to-date and will also allow us to continue our research projects as planned. Multicolor sorts at the Universitätsklinik have become prevalent in recent years, but the current sorter has neither the number of PMTs nor the optical sensitivity required to be able to see or differentiate many of the populations which need to be sorted, especially in the far-red region of the spectrum. In addition, the current sorter does not have all of the lasers which we require, for example, there is no 561 nm laser for the excitation of mCherry. Furthermore, many of the samples which need to be sorted are from primary human samples and the current machine does not have any active operator protection. A replacement cell sorter with a biosafety hood would provide the best operator protection, as well as protecting others present in the room. The replacement cell sorter would be located in a core facility setting so that groups from the entire Uniklinik would have access to it, as has been the case for the current cell sorter over the last 17 years.
