Analytical ultracentrifugation (AUC) is a powerful tool to characterize proteins and protein complexes.Proteins are analyzed in solution without a supporting matrix. AUC allows for the accurate determination of protein native molecular mass, complex subunit stoichiometry, complex assembly and associations, as well as protein conformation and shape. AUC requires little material compared to other methods and allows characterization of proteins (complexes) over a wide range of molecular weights and association constants.The machine applied offers a double detection system using absorbance and Rayleigh interference optics. Interference optics allow for rapid scan speeds and produce much more detailed data than absorbance optics alone. They also allow for the accurate detection of concentrated as well as non-absorbing samples.
