It is still largely unclear which membrane protein degradation mechanisms operate locally at synapses, despite their importance for health and diseases. Project A17 is devised to characterize the protein scaffolds responsible for local membrane protein sorting and degradation. We have identified five novel players at axon terminals: four neuronal Rab GTPases (Rab19, Rab26, RabX1 and RabX4) and at the multiple-protein interacting coiled-coil protein and RabGAP (Evi5). We seek to understand the dynamics and interaction network of these proteins and their role in the turnover of membrane-associated proteins, including the active zone scaffold Brp, at axon terminals.
