The sulfonyl group (-SO2-) is one of the most abundant structural motifs in medicinal chemistry. Currently there are over 100 approved drugs containing a sulfon, sulfonamide or sulfonic ester functional group. Since 80 years molecules with a sulfonamide functionality belong to the most important drugs. Among the 200 most prescribed drugs in the US in 2012, close to 10% bear a sulfonamide group. This project is focused on the development of novel procedures for the preparation of sulfonamides. Our goal are consistent and practical methods for the modular synthesis of a wide variety of structurally diverse sulfonamides. Therefore several complementary multicomponent reactions will be developed. All methods will be based on the installation of the central structural motif, the sulfonyl functionality, via sulfur dioxide.
