Non-coding RNAs as barriers in emerging viral infections

Non-coding RNAs play a major regulatory roles during viral infection of host cells. However, with the notable exception of miRNAs, not much is known about (long) ncRNAs during viral infection.Here, we will annotate ncRNAs in main virus reservoirs, such as camelids, rodents, bats and insects.In this proposal we aim to systematically understanding the role of regulatory RNAs to define species barriers in emerging viral diseases. We use a three-dimensional approach to tackle this timely question. The three axes are virus, host and tissue. First, we will investigate a constant virus-host- systems and vary the tissue axis. By differentially expression analysis we aim to understand tissue specificity. In a second approach we will choose constant host-tissue-systems and infect them with various viruses. With this approach we will be able to discriminate general ncRNAs, ncRNAs during common viral infections and ncRNAs being specific for a virus or a viral family. In a third approach we will keep the virus and the infected tissue constant and vary along the host axis. With this approach we aim to identify species barriers at a host level. We will collect data generated by RNA-Seq within the priority programme, which are synchronized with the same RNA extraction and library preparation protocols, and complement them with a few settings in order to fulfill the main aim to understand species barriers in emerging diseases.Additionally, we will provide a bioinformatical core-facility to the members of the priority programme.
