We study a Type 1 secretion system (T1SS), which translocates the human pathogenic RTX toxin HlyA in one step across both membranes of E. coli. The project relates the dynamic assembly of the translocation complex with the molecular identity of the transport-competent components of this machinery. In a combined approach of in vitro and in vivo experiments the following key aspects will be addressed: (1) identification of switches and triggers involved in identity changes in T1SS (structural studies), (2) addressing the molecular interaction network and in vitro reconstitution of a T1SS (functional studies) and (3) analysis of cellular HlyA secretion and a detailed in vivo / in vitro correlation (cellular studies).
