Endosome-coupled mRNA transport is essential for efficient hyphal growth and unconventional secretion in Ustilago maydis. Here, we address firstly how higher-order mRNP complexes are attached to shuttling endosomes. To this end we will perform a structure/function analysis of the key endosomal RNA-binding protein as well as identify novel endosomal interaction partners involved in mRNP attachment. Secondly, we will carry out a structure/function analysis of a novel anchor protein for unconventional secretion as well as identify new candidates by proximity labeling that carry out a specific function in the zone of un-conventional secretion.
