Although autophagy is one of the fundamental cellular protein degradation mechanisms, the regulation of autophagosomal membrane compartments is still not fully understood. Using state-of-the-art NMR spectroscopy in concert with other biophysical and biochemical techniques we will investigate the conformational dynamics and functional mechanism of GABARAP, a key autophagy-related protein which alternates between a soluble form in the cytosol and a membrane-anchored form to mediate membrane tethering and hemifusion during autophagosome biogenesis.
