Zusammenfassung der Projektergebnisse
Das Zentrum für Regenerative Therapien Dresden (CRTD) hat führende Wissenschaftler aus den Bereichen Entwicklungs-, Regenerations- und Stammzellbiologie vereint, um die Forschung zu Geweberegeneration und Zellersatz voranzutreiben. Das Prinzip des CRTD, die Entdeckung aus erfolgreicher Regeneration und Entwicklung, basiert auf außergewöhnlicher Forschung an Stammzellen und mit Modellorganismen, die ihre Fähigkeit zur Regeneration von Geweben nach einer Schädigung bewahren. Ein effektives Merkmal des CRTD ist die wissenschaftliche Vielfalt der Modelle, Organe, Krankheiten und Ansätze, die durch die gemeinsamen Ziele und die Nähe von Wissenschaftlern und Klinikern synergetisch genutzt werden. Mit der Förderung konnte ein Netzwerk aufgebaut werden, das die Grundlagenforschung der bestehenden Technische Universität Dresden (TU Dresden) Forschungsinstitute mit klinischen Instituten und mit außeruniversitären Instituten eng verbindet. Der Einsatz von Seed Grants und Gerok- Positionen schuf den Rahmen für die interdisziplinäre Verzahnung. Der Ausbau und das gezielte Schließen von Lücken im Großgerätebereich in den Technologieplattformen und im Service- und Gerätepool hat dem Campus einen Vorteil verschafft. Innerhalb des Clusters konnten neue Ansätze etabliert werden, die dann auf den Campus ausgedehnt wurden, wie z.B. Tenure Track Optionen oder flexible Kinderbetreuung. Die Grundlagenforschung im Bereich regenerative Modellorganismen des CRTD konnte international renommierte Forscher der Zebrafisch- und Salamanderforschung sowie Experten für Säugetiermodelle integrieren. Die Forscher haben bahnbrechende Entdeckungen gemacht, die neue Forschungswege eröffnen und die Dresdener Forschung prägt. Die Zelltherapie ist ein weiteres zentrales Forschungsgebiet. Um dieses strategische Ziel des CRTD zu erreichen, wurde in Dresden die Inseltransplantation der Bauchspeicheldrüse erstmalig in Deutschland etabliert. Diese Fortschritte bieten auch Möglichkeiten für die Transplantation von hESC- und hiPSC-basierten Beta-Zellen. Zell-, organoid- und in vivo basierte Methoden zur Pathogenese und Identifizierung von Wirkstoffzielen wurden durch exzellente Dresdner Entdeckungen und Rekrutierung entwickelt. Eine Grundlage für die Entdeckung pathogenetischer Mechanismen und des Wirkstoffscreenings wurde durch in vivo Zebrafisch-Drug-Screening geschaffen. Die Entdeckungen, die für hämatopoetische Stamm- und Vorläuferzellen (HSPC) und die pankreatische Beta-Zell-Expansion relevant sind, ermöglichen eine erfolgreiche iPSC/ESC/ gene editing Facility. Es gab auch therapeutisch relevante zellbasierte Entdeckungen bei Kindern mit immunologischen Erkrankungen, darunter bahnbrechende Geburtskohorten für Typ-1-Diabetes und Immunität sowie technologische Entwicklungen mit Relevanz für Fische über Mäuse bis hin zum Menschen. Der Freistaat Sachsen und die TU Dresden haben das CRTD mit einer jährlichen Förderung von derzeit 9,9 Mio. € dauerhaft übernommen. Das CRTD ist als eines von drei Instituten innerhalb des Center for Molecular and Cell Bioengineering (CMCB) organisiert, einer fakultätsähnlichen Verwaltungsplattform für CRTD, B CUBE und BIOTEC. Das CRTD wird somit weiterhin seine wissenschaftliche Mission verfolgen und seine Arbeit im Bereich der Regenerativen Biologie - und Gewebeforschung fortsetzen können.
