The project D4 (Höfer/Freund) integrates experimentation and modeling to elucidate how phosphorylations of scaffolding proteins cause the reversible assembly of a module that regulates T cell adhesion. The interactions of the scaffolds ADAP and Vav with the Src-family kinase Fyn and the phosphatase SHP-2 as well as the resulting phosphorylation dynamics are studied in a reconstituted system and in the cellular context. The modeling makes testable predictions on the role of competing feedback loops for kinase and phosphatase activation and the control exerted on signal transduction by individual protein binding and phosphorylation steps.
