Membrane translocation and chaperone-dependent folding of the lipase A of Pseudomonas aeruginosa (A10#)

Subject Area Biochemistry
Term from 2018 to 2023
Project identifier Deutsche Forschungsgemeinschaft (DFG) - Project number 267205415

Extracellular lipases are recognized as virulence factors of Pseudomonas aeruginosa, however their chap-erone-mediated folding at the lipid membrane interface is not understood. Here, we set out to elucidate how the membrane environment steers lipase biogenesis. We will employ the newly reconstituted system and in vivo experiments to study (i) how the membrane-anchored LipH chaperone contributes to the lipase A translocation, (ii) whether lipids or membrane-associated proteins regulate the lipase:LipH assembly, and (iii) how the lipase folding is influenced by the crowded and dynamic environment at the membrane interface.
