The membrane-permeable derivatives of the second messengers NAADP, ADPR and cNMPs (MASTER-compounds) developed in the 2nd funding period will be further modified by new masking groups as well as photocleavable groups for spatial and temporal resolution of the biological processes. These biochemical tools will be used for cell-based studies to understand their biological role, e.g. NAADP/Ca2+ signaling in T cell aging. All-in-one-clickable derivatives will be prepared to identify intracellular location and expression of NAADP receptors. NAADP antagonists will be addressed as well.
