The project deals with the role of lysosomal proteases in Parkinson’s disease, which is characterized by conversion of α-synuclein from a soluble synaptic protein into insoluble amyloid fibrils accompanied by degeneration of dopaminergic neurons. The interplay of α-synuclein with lysosomal cathepsins D, B and L, involved in lysosomal degradation of α-synuclein, will be studied. Using iPS cell derived dopaminergic neurons, patient mutations in cathepsins will be examined, and cathepsins D, B and L will be analyzed as therapeutic targets in Parkinson’s disease.
