The project studies the role of matrix metalloproteases (MMPs) in the proteolytic invasion of macrophages. Within cells, MMPs travel bidirectionally along microtubules, regulated by specific kinesin and dynein motor proteins and members of the RabGTPase family. A novel kinesin regulator of MMP-dependent extracellular matrix proteolysis was discovered. The role of this kinesin in the recycling pathways of MT1-MMP will be studied. Inhibitory peptides are developed and tested for modulation of macrophage invasion in a pathophysiologically relevant context.
