We have found that two DNA glycosylases, NEIL1 and NEIL2, coordinate nucleobase processing during TET/TDG-dependent DNA demethylation. To investigate the physiological relevance and bio-logical role of Neil DNA glycosylases in DNA demethylation we have generated mice as well as mouse embryonic stem cells mutant for Neil genes. The aim of this project is to characterise these mutant cells for their differentiation capacity and their profiles of oxidized cytosine, and to assess potential developmental defects of Neil mutant mice.
