This project aims at elucidating how the DNA replication machinery reacts to DNA lesions. We have gener-ated a large collection of engineered human and mouse cells with labelled components of the replication machinery as well as sensors for ubiquitin modifications in live cells. Using these models and a combination of microscopical and genomics approaches, we will generate functional and kinetic spatio-temporal models of the response of the genome replication machinery to damages and how cellular physiology is affected.
