Excessive, unregulated macrophage activation is linked to disease development in haemophagocytic lymphohistiocytosis/macrophage activation syndrome (HLH/MAS). Macrophage activation is regulated metabolically, and inhibition of metabolic processes linked to activation offers an opportunity to control HLH/MAS. In this context, the project has two aims: 1) To characterize the metabolism, as related to function, of macrophages from a mouse model of HLH and from HLH/MAS patients, and 2) To block macrophage activation in HLH/MAS using small molecule inhibitors of key metabolic enzymes.
