Project C06 will map the transcriptome during resolution of inflammation in arthritis, thereby deciphering the responsible cellular interactions at the molecular level. Previous funding periods have identified several cell types that promote resolution in arthritis (Tregs, ILC2, rEOS). To fulfil their task, these cells require a specific microenvironment of stromal cells. Our data show that regionally distinct tissue microenvironments exist that form either pro- or anti-inflammatory milieus. The project aims to identify the cellular interactions in these respective stromal environments and validate them as therapeutic targets. Furthermore, based on the transcription maps, an algorithm for computer-assisted decision of therapy selection will be developed.
