High Resolution Confocal Spinning Disc System

Spinning disk confocal (SDC) microscopy is one of the most successful imaging methods to investigate subcellular dynamics. The fast and sensitive camera based acquisition combined with a confocal mode enable researchers to perform multicolor 3D live-cell experiments over extended times. The AMBIO core facility currently has only one SDC system that has been successfully used in ~ 80 projects from research groups throughout Berlin. Not surprisingly, this system is frequently overbooked, limiting the planning of live-cell experiments. The lack of advanced features further prevents several groups from performing optimized imaging projects. Several high-end user groups (i.e. AGs Reber, Harms, Eickholt, Tacke, s. Supplement for Research Groups) who requested more usage capacities and advanced technical features are therefore part of this proposal to procure a novel SDC system that in addition to previous functionalities (multiplexing, automation) offers novel features (i.e. large field of view, optics for tissue imaging, super resolution) that enhance throughput and resolution. Beyond these groups the proposed system will be available to all AMBIO users (currently ~ 115 from ~ 42 research groups) to perform improved high-resolution live-cell imaging projects.
