Nanoscale regulation of molecular switches at the Golgi (A20)

Project A20 will investigate the mechanisms of regulation of ARF GTPases at the ER-Golgi interface. In light of their findings suggesting a differential role for ARF1 and ARF4 in ER-Golgi transport and organization, during the next funding period A20 will address how the two GTPases are recruited to different membrane nano-domains and activated by ARF GDP-GTP exchange factors (GEFs). They will employ CRISPR-Cas9 knock-in methodology in combination with proximity biotinylation using TurboID and mass spectrometric analysis, dynamic confocal and live-cell STED microscopy and perturbation via CRISPR-Cas9 knock outs of ARF GEFs.
