Atomic Force Microscope

We apply for an atomic force microscope, which will be used to investigate relationships between the structure, mechanics and function of biomolecular complexes. We will study the topology and viscoelastic properties of whole cells, for examle after application of an external force. Furthermore, cellular components, in particular cytoskeletal filaments, will be investigated with regard to molecular structure and force-distance behaviour under tension. A large travel range in lateral (x-y-plane) as well as vertical (z) direction is essential for the experiments proposed here, since the objects under investigation may be several micrometers large. At the same time, a high spatial resolution is required to image substructures in the nanometer range. Since we will primarily image biological samples, the measurements will be performed in liquid.
