The project investigates the interaction of the SARS-CoV-2 macrodomain (Mac1) with the host antiviral response. It uses newly identified, specific small-molecule inhibitors. The lead (pro)drug will be optimised and its pharmacokinetic, pharmacodynamic and toxicological properties evaluated in vivo. The inhibitor’s activity against related viral macrodomains will be assessed. AAV-delivered anti-Mac1 intrabodies will be developed as diagnostic and inhibitory tools. Host interaction partners of Mac1 will be identified to clarify the molecular basis of Mac1-mediated host response suppression.
