In vivo imaging of distinct leukocyte populations on multiple scales is vital for understanding their dynamics and roles in inflammation. To avoid premature label loss of ex vivo labelled cells, e.g., ER-HoxB8, we have devised self-labelling enzyme or nanobody (Nb) epitope tags to be genetically introduced as modular tagging platforms. These tags can be visualized in vivo days after cell differentiation and re-injection with label-customized substrates or Nbs. Focusing on radioactive labels as robust readouts in whole-body imaging, we plan to enhance the signal-to-background ratio by using improved and engineering covalently binding Nbs and by increasing their specific radioactivity through novel chemical separation methods.
