Myeloid cell subtype-specific functions in immune complex-driven nephritis

Immune complexes (IC)-mediated pathologies (also known as type III hypersensitivity reactions) affect millions worldwide, with a very high socioeconomic burden. Yet, the mechanisms of sIC-mediated inflammation remain poorly defined. Our recent work has provided unique compelling evidences that the macrophages that resident in the renal interstiti-um, termed kidney-resident macrophages (krM omega s), scavenge sIC and initiate the inflammatory response to circulating sIC in the kidney. However, it is unclear how this inflammatory response triggers tissue injury and if tissue injury is mediated by macrophages, or other infiltrating myeloid cells. In this proposal, we will systematically interrogate the sequence of events that lead to the path-ogenesis of type III hypersensitivity disease in the kidney and the role of myelid cells in nephritis.
