In summary, our contribution to different registries were of high relevance during the pandemic. REV and KW provided important information to patients and regularly updated recommendations for the treatment of patients with inflammatory rheumatic diseases or primary immunodeficiency and the biobanked samples were provided for several research projects. We addressed the vaccination response in healthy individuals. HE revealed that the memory B cell response after 3-4 vaccinations with SARS-CoV-2 mRNA vaccines was long lasting and that stable antibody titers prevent the binding of Spike protein to the receptor ACE2. Our analysis of patients with immunodeficiency due to impaired immunoglobulin production revealed that these patients are prone to have a prolonged disease course. CD4 T cell responses were enhanced while CD8 responses were compromised. Despite an impaired memory formation antigen-specific B cells were present after infection with SARS-CoV-2, though not showing the differentiation state as observed in immunocompetent controls. Unlike in immunocompetent individuals these patients benefited from convalescent plasma. Lastly, our studies contributed to deepen the understanding of the enigmatic CD21low B cell population which is not only expanded in the peripheral blood of CVID patients. Analyzing this subset in the antigen-specific immune response of immunocompetent individuals to SARS-CoV-2 revealed that they can arise from any other B cell population, which was not clear till now. Additionally we showed that these enrichment of these cells may predict a poorer outcome of SARS-CoV-2 vaccination.
