Westermann will use a combination of biochemical reconstitution and high-end imaging to de-fine the molecular mechanisms that establish kinetochore bi-orientation and thereby trigger the metaphase-anaphase cell state transition in the model system Saccharomyces cerevisiae. The project aims to 1) define the molecular requirements for the assembly of a plus-end associated outer kinetochore, 2) reveal the molecular mechanisms of Mps1 recruitment to the outer ki-netochore and analyse its contribution to bi-orientation and 3) analyse outer kinetochore as-sembly and lateral to end-on conversion at dynamic plus-ends in vitro.
