Boos will investigate the molecular mechanisms and regulation of pre-initiation complex for-mation, the key determinant for origin firing and thus initiation of DNA replication. His project will address (1) how the pre-IC is assembled on the molecular level, and (2) how this assembly is regulated by cell cycle, DNA damage checkpoint and other kinases. He will use a hybrid reconstitution system based on frog egg extracts and purified components to monitor in situ assembly using sophisticated mass spectrometry approaches. Moreover, he will use chemical approaches to define how the process is controlled by kinases.
