Nanovaccines accumulate in the liver typically in tolerance-promoting KC (Kupffer cells) and LSEC (sinusoi-dal liver endothelial cells). Both cell types can be reprogrammed to serve as antigen presenting cells by applying suitable adjuvants. B15N develops nanovaccines which besides dendritic cells also address and activate KC/LSEC. This approach serves to induce potent anti-tumor responses in liver and skin (metastatic melanoma). The suitability of protein nanocapsules containing peptid antigen/adjuvant and of liposomes loaded with antigen-coding mRNA/nucleic acid-based adjuvants to serve as nanovaccines will be evaluated.
