The proposed instrument is an integrated system consisting of a sealed tube X-ray generator, double focusing mirror optics, diffractometer, modern pixel array detector, cryo system, stage for in situ screening of crystals and miscellaneous items. The system will also guarantee the integration of an existing Arinax HC-Lab dehumidifier with manual nozzle exchanger. The proposed diffractometer will replace an existing system which was bought with funds from the Deutsche Forschungsgemeinschaft via funding to the Rudolf Virchow Zentrum für Experimentelle Biomedizin (FZ-82) in 2005 and was installed in 2006. The existing unit will have reached an advanced age of 15 years at the time of replacement and neither no longer represents current technology nor operates in a reliable manner. In particular, the rotating anode generator is plagued by significant down times due to problems with the vacuum and anode. Even when operational, the intensity is significantly reduced due to aging of the mirror system and the detector technology (imaging plate) is two generations behind the current hybrid pixel array detectors and suffers from low sensitivity, high background noise and excessive readout time. Replacing the existing diffractometer, the only system for macromolecular crystallography in Würzburg, is essential to provide the researchers listed in this proposal, who belong to the Rudolf Virchow Center, a central entity of the University of Würzburg and three different departments (Biology, Chemistry and Pharmacy as well as Medicine) access to a state-of-the-art in-house X-ray diffractometer.
