HIV-infection cannot be cured as the virus persists during treatment as integrated latent provirus. However, reduction of the latent reservoir could be achieved by reversing latency, so that infected cells can be elimi-nated by the immune response. We will use a library of hundreds of cell lines containing latent HIV provirus-es based on a latency reporter system combined with genetic, biochemical and microscopy techniques to identify the determinants of HIV latency reversal. This project will provide a better understanding of the role of the integration context for HIV latency and the mechanisms underlying latency reversal.
