The nuclear Ras-like GTPase Ran has been implicated in a wide variety of cellular processes, including nucleocytoplasmic transport, cell cycle progression, maintenance of chromatin and nuclear organization, nucleolar structure, DNA replication, chromosome segregation, RNA metabolism, microtubule organization, and yeast mating. A direct function for Ran and some of its interacting factors, however, has so far only been established for nucleocytoplasmic transport. It is therefore unclear whether the diversity of processes affected by Ran and its associated factors is a indirect consequence of their primary role in nucleocytoplasmic transport, or is due to multiple functions of Ran in the cell. In order to reveal the molecular basis of Ran's functional diversity we are analyzing the cellular function of Ran-interacting proteins in the yeast Saccharomyces cerevisiae. Besides a functional analysis of established or suggested Ran-interacting proteins, we aim at the identification and characterization of novel representatives of such proteins in this organism.
