Angiogenesis, the sprouting of new capillaries from pre-existing blood vessels, is a process that is subdivided into the following components: proteolytic degradation of extracellular matrix, migration of endothelial cells toward a chemotactic stimulus, proliferation at the base of the new sprout and maturation of the vessel by recruitment of perivascular cells and deposition of new matrix. The focus of the project described in the present application is to investigate the roles of the Angiopoietin/Tie-2 and the ephrinB/EphB ligand/receptor systems for capillary sprouting, during which both types of systems have no direct impact on the regulation of cell proliferation. Based on our in vitro findings gathered during the previous application period, the present questions are as follows: the role of the induction of proteases for the Angiopoietin-induced hypervascularization in vivo, the identification of signal transduction molecules that are important for Angiopoietin and ephrinB mediated endothelial cell sprouting, the investigation of the possible role of the Tie-1 receptor as a modulator of Tie-2 activity, and potential functional interactions of the respective ligand/receptor systems.
