Mammalian gene expression patterns are regulated by enhancers. To activate genes, enhancers interact with gene promoters in specific 3D chromatin structures. The mechanistic basis of the formation of these structures and enhancer-mediated gene activation is not known. The Mediator complex is thought to play a central role in the transmission of activating signals from enhancers to gene promoters, but its role in the formation of enhancer-promoter interactions remains unclear. The aim of this project is to elucidate the molecular basis of the function of the Mediator complex in regulating enhancer-promoter communication and gene expression patterns during cellular differentiation.
