A04 aims to uncover the role of mechanics in regulating human neural stem cell lineage decisions, neuron formation and neuronal migration leveraging 3D brain organoids. We will use engineered matrices to quantitatively tune the physical properties of the brain organoid environment and employ time-lapse imaging to follow single cells and their progeny thereby quantifying lineage decisions and their trajectories in a tissue-like context. To uncover the molecular framework relaying changes in the physical environment to alterations in cellular behaviour, we will use single cell RNA-sequencing and functionally assess associated key nodes in developmental neurogenesis.
