The generation of tissue Treg cells is a stepwise process, and we could recently demonstrate that this is accompanied by extensive epigenome remodeling, finally leading to the acquisition of unique tissue-specific epigenetic signatures. We here aim to characterize the precise dynamics of tissue-seeding and epigenetic remodeling of murine and human tissue Treg cells, and unravel the signals and molecular mechanisms by which these cells acquire and stabilize their unique functional programs. These findings will contribute to the generation of a new class of tissue-specific and functionally adapted Treg cells for future therapeutic applications.
